Bioisosteric modification on melatonin: synthesis of new naphthalene derivatives, in vitro antioxidant activity and cytotoxicity studies BJPS
Shirinzadeh,Hanif; Ghalia,Mohammed; Tascioglu,Alev; Adjali,Ferial Intissar; Gunesacar,Gulsen; Gurer-Orhan,Hande; Suzen,Sibel.
Melatonin (MLT) is a strong free radical scavenger that protects the body from the deleterious effects of excess oxidants. Synthesis of MLT analogue compounds with antioxidant potency has recently attracted the interest of researchers. In general, the strategy consists of modifying the groups in the different sites of the indole ring or replacing the indole ring with an analogue. As part of our ongoing research, the antioxidant capacity and cytotoxicity of newly synthesized MLT analogue naphthalene derivatives were evaluated. The radical scavenging activity was tested by a 1,1-diphenyl-2-picrylhydrazyl (DPPH) assay. Most of the synthesized compounds showed significant antioxidant activity in comparison to MLT. The structure-activity relationship was...

Bovine serum albumin-based nanoparticles containing the flavonoid rutin produced by nano spray drying BJPS
Pedrozo,Regiellen Cristina; Antônio,Emilli; Khalil,Najeh Maissar; Mainardes,Rubiana Mara.
Rutin is a flavonoid compound obtained from different vegetables and fruits; specifically, it is found in the seeds of buckwheat and in fruit peels, particularly citrus. It is also an important constituent of red wine. Rutin exhibits various biological properties including antiviral, vasoprotective, anti-inflammatory, and anticarcinogenic activities. However, its antioxidant activity is the most well studied. Despite the potential for in vitro applications, rutin presents low oral bioavailability that affects its biological activities. Nanoparticles composed of polymers, protein, or lipids are of great importance in the pharmaceutical and nutraceutical areas due to their physicochemical properties, which improve the pharmacokinetics of the drug which is...

Brazilian propolis protects Saccharomyces cerevisiae cells against oxidative stress BJM
Sá,Rafael A. de; Castro,Frederico A.V. de; Eleutherio,Elis C.A.; Souza,Raquel M. de; Silva,Joaquim F.M. da; Pereira,Marcos D..
Propolis is a natural product widely used for humans. Due to its complex composition, a number of applications (antimicrobial, antiinflammatory, anesthetic, cytostatic and antioxidant) have been attributed to this substance. Using Saccharomyces cerevisiae as a eukaryotic model we investigated the mechanisms underlying the antioxidant effect of propolis from Guarapari against oxidative stress. Submitting a wild type (BY4741) and antioxidant deficient strains (ctt1∆, sod1∆, gsh1∆, gtt1∆ and gtt2∆) either to 15 mM menadione or to 2 mM hydrogen peroxide during 60 min, we observed that all strains, except the mutant sod1∆, acquired tolerance when previously treated with 25 µg/mL of alcoholic propolis extract. Such a treatment reduced the levels of ROS...

Cadmium-Zinc interactions in a hydroponic system using Ceratophyllum demersum L.: adaptive ecophysiology, biochemistry and molecular toxicology Braz. J. Plant Physiol.
Aravind,Parameswaran; Prasad,Majeti Narasimba Vara.
The interaction between an essential micronutrient, Zn and a toxic non-essential element, Cd has been comprehensively reviewed based on our experiments conducted with Ceratophyllum demersum L. in a hydroponic system. Since Cd and Zn belong to the group IIB transition elements and show similarities in chemistry, geochemistry and environmental properties, it would be one of the elemental pairs of choice to investigate metal-metal interactions. Evidence in support of the protective role of Zn against Cd toxicity in Ceratophyllum demersum L. is presented in this review. Based on our experimental results, we conclude that the antioxidant properties of Zn play an important role in counteracting Cd toxicity.

Changes in fatty acid composition in the giant clam Tridacna maxima in response to thermal stress ArchiMer
Dubousquet, Vaimiti; Gros, Emmanuelle; Berteaux-lecellier, Veronique; Viguier, Bruno; Raharivelomanana, Phila; Bertrand, Cédric; Lecellier, Gael J..
Temperature can modify membrane fluidity and thus affects cellular functions and physiological activities. This study examines lipid remodelling in the marine symbiotic organism, Tridacna maxima, during a time series of induced thermal stress, with an emphasis on the morphology of their symbiont Symbiodinium. First, we show that the French Polynesian giant clams harbour an important proportion of saturated fatty acids (SFA), which reflects their tropical location. Second, in contrast to most marine organisms, the total lipid content in giant clams remained constant under stress, though some changes in their composition were shown. Third, the stress-induced changes in fatty acid (FA) diversity were accompanied by an upregulation of genes involved in lipids...

Chemical, enzymatic and cellular antioxidant activity studies of Agaricus blazei Murrill Anais da ABC (AABC)
HAKIME-SILVA,RICARDO A.; VELLOSA,JOSÉ C.R.; KHALIL,NAJEH M.; KHALIL,OMAR A.K.; BRUNETTI,IGUATEMY L.; OLIVEIRA,OLGA M.M.F..
Mushrooms possess nutritional and medicinal properties that have long been used for human health preservation and that have been considered by researchers as possible sources of free radical scavengers. In this work, the antioxidant properties of water extracts from Agaricus blazei Murill, produced by maceration and decoction, are demonstrated in vitro. Resistance to oxidation is demonstrated through three mechanisms: i) inhibition of enzymatic oxidative process, with 100% inhibition of HRP (horseradish peroxidase) and MPO (myeloperoxidase); ii) inhibition of cellular oxidative stress, with 80% inhibition of the oxidative burst of polymorphonuclear neutrophils (PMNs); and iii) direct action over reactive species, with 62% and 87% suppression of HOCl and...

Circadian rhythms of resistance to UV-C and UV-B radiation in Euglena as related to 'escape from light' and 'resistance to light' OAK
Bolige, Aoen; Kiyota, Maki; Goto, Ken; 後藤, 健.
Radiation-induced stress, either from visible or UV light, is strongest at midday. We found that, in the absence of stress or time cues, Euglena gracilis Z was the most resistant to UV-C and UV-B at subjective midday, whether judged from immediate or reproductive survival. The circadian UV-resistance rhythms were free-running in stationary cultures under 1-h light/1-h dark cycles or continuous darkness, indicating that cell-cycle dependent DNA susceptibility to UV was not involved. We moreover examined what was the primary cause of the circadian U-V resistance, estimated as the immediate cell survival. The half-maximal lethal dose (LD50) of UV-C at subjective midday (the most resistant phase) was 156 J/m(2), which is similar to 3-fold that at subjective...

Circadian rhythms of the L-ascorbic acid level in Euglena and spinach OAK
Kiyota, Maki; Numayama, Naoko; Goto, Ken.
Plant defenses against photo-oxidative stress have been studied almost exclusively with respect to stress responses, and little is known about how non-enzymic antioxidants change under constant conditions without a time cue or an environmental stress. Here, we show that, in both the flagellated alga Euglena gracilis Z and the angiosperm Spinacia oleracea L., the potent antioxidant L-ascorbic acid (Asc) displays a circadian rhythm with a maximum at subjective midday, a physiological state reflecting that attained at noon under daily light/dark cycles. Thus, photosynthetic organisms can maximize antioxidant levels in anticipation of midday, when photo-oxidative stress is most severe. These results may partly explain the in-phase circadian UV-C resistance...

Coenzyme Q10 and its effects in the treatment of neurodegenerative diseases BJPS
Santos,Graciela Cristina dos; Antunes,Lusânia Maria Greggi; Santos,Antonio Cardozo dos; Bianchi,Maria de Lourdes Pires.
According to clinical and pre-clinical studies, oxidative stress and its consequences may be the cause or, at least, a contributing factor, to a large number of neurodegenerative diseases. These diseases include common and debilitating disorders, characterized by progressive and irreversible loss of neurons in specific regions of the brain. The most common neurodegenerative diseases are Parkinson's disease, Huntington's disease, Alzheimer's disease and amyotrophic lateral sclerosis. Coenzyme Q10 (CoQ10) has been extensively studied since its discovery in 1957. It is a component of the electron transportation chain and participates in aerobic cellular respiration, generating energy in the form of adenosine triphosphate (ATP). The property of CoQ10 to act as...
